Biography

Anton Wagenmakers is a world-leading (4*) researcher according to UK Research Excellence Framework [REF] criteria) with 190 peer reviewed publications in SCI/SSCI journals and an h-index of 54 (Web of Science) and 70 (Google Scholar). His research is focussing on 1) mechanisms by which a sedentary lifestyle via the resultant impairments in fat metabolism in skeletal muscle fibers and adipose tissue cells leads to impaired glucose tolerance, insulin resistance, diabetes and sarcopenia in obese and elderly individuals and on 2) the design of optimal exercise/nutrition interventions, that can be implemented in real life, and are able to prevent, postpone or reverse the development of the ageing phenotype and chronic age and lifestyle related diseases. In the intervention studies, Anton investigates the underlying molecular and physiological mechanisms using expertise in methods to measure glucose, fatty acid and protein metabolism in living human beings with stable isotope tracer methods and state-of-the-art confocal immunofluorescence microscopy methods. The latter stain key enzymes playing a role in insulin resistance mechanisms in sections of skeletal muscle to thus visualise the spatial distribution of key enzymes in glucose, fatty acid and lipid metabolism to include their co-localisation with the plasma membrane, mitochondria and lipid droplets. Other novel methods stain endothelial enzymes controlling transendothelial transport of glucose and fatty acids and production of the vasodilator NO specifically in the endothelial layer of skeletal muscle capillaries and arterioles. This research has generated compelling evidence that insulin resistance occurs both in skeletal muscle fibers and their microvasculature. In obesity, metabolic syndrome and ageing insulin mediated dilation of skeletal muscle arterioles is impaired due to failure of insulin to activate eNOS, the enzyme producing NO. As result muscle fibres in well-fed bodies are starved of nutrients and meal derived glucose, lipids and amino acids are deposited in non-muscular tissues such as liver and adipose tissue leading to gradual downward spiral towards increased adiposity, insulin resistance, glucose intolerance, diabetes, sarcopenia and neurodegeneration in obese elderly individuals.

Anton is academic lead of the Exercise Metabolism and Adaptation Research Group (EMARG). With 20 members of academic staff, the majority being early and mid-career, EMARG is the largest research group in RISES. EMARG staff is covering a large range of research activities ranging from Sport Performance to Exercise for Health and Prevention of Chronic Diseases and Development of the Ageing Phenotype covering the full range from fundamental to applied research and from the molecular level to whole body physiology.

Anton as first or senior supervisor has led 10 PhD students in the Netherlands and 15 PhD students in the United Kingdom to a successful completion of their PhD. Four of his former PhD students are full time Professor in Sweden, Denmark, the Netherlands, and UK. All have an appointment in academia or Life Science companies.
